The roadside bombing took place in Trinkot city in the province of Uruzgan, as the police officers were patrolling the area. A sixth officer was injured in the incident as well.
Although no group has yet claimed responsibility for the attack, Afghan officials hold Taliban militants accountable.
The province of Uruzgan shares borders with Kandahar and Helmand, which are considered to be under Taliban influence.
On Friday, a US-led airstrike killed three in central Afghanistan.
Although the US-led invasion of Afghanistan in 2001 removed the Taliban from power, insecurity continues to rise across the country, despite the presence of some 130,000 US-led troops.
More than ten years into the war, Washington has now turned to negotiations with the Taliban militants.
