Protesters shouted slogans against the US on Thursday, threatening to block the Torkham-Jalalabad road unless the prisoners are freed within 48-hours.
The three civilians were arrested by the American forces in Ghani Khel district last month.
There had been no news on their whereabouts until recently, when it became clear that they had been taken to the Bagram Airbase north of Kabul, where a large number of Afghans are held without charge or trial.
President Hamid Karzai has demanded the US hand over control of the American-run prison to Afghan authorities, saying that keeping Afghans there violates the country’s constitution.
The US-led invasion of Afghanistan was launched in 2001. The offensives removed the Taliban from power, but insecurity continues to rise across the country.
The growing number of military casualties in Afghanistan has intensified opposition in the United States and other NATO member states against the protracted war in the country.
